Current porous resins entrap biodrugs and contaminants and require large volumes of buffers for purification. When large volumes of initial, raw broth have to be processed, the time to perform these purifications increases dramatically. Orbicell® bioresins from APL selectively bind target biodrugs on the surface and do not retain or entrap contaminants. The nonporous character of the bioresins allows for a rapid removal of unbound contaminants. APL's process reduces the number of steps to produce final product and does so in a fraction of the time compared to traditional column chromatography. Manufacturers using chromatographic columns for large-scale purification view the validation procedure as the most critical and problematic aspect of the total purification process. In comparison to packed columns Orbicell® bioresins can be cleaned and validated in a fraction of the time. Almost all of the ligands used for attachment onto Orbicell® bioresins can be safely autoclaved or sterilized with NaOH, thus minimizing the problems with validation. |

In the old, classic column system, purification solutions will seek out the most unobstructed route through the column; thus "channeling" occurs. Channeling leads to co-elution of product and contaminants with the column. Moreover, low flow regions of the column will tend to build-up contaminants that elute with the target biodrugs during purification. |
